Employees at the Greyhound station called police after seeing the man with a gun at around 11 a.m.
As police responded, the man held himself up inside a bathroom on an empty bus.
Officers heard a single gunshot and called in SWAT, who responded and found that the man had shot himself inside the bathroom.
The fire department was called in open the door to the bathroom after it got jammed. A doctor on scene pronounced the man dead.
Parts of the station were evacuated and no other injuries were reported.
Police have not released the name of the gunman.
A Greyhound spokeswoman says the Cleveland-to-Dallas bus had emptied at a scheduled stop in Cincinnati and that the man was among passengers who were told by a bus company employee to move their smoking to another area.
Several roads around the bus station were closed during the investigation. The ramp to Gilbert Ave. from SB 71 was also closed, but has since reopened.
